Crl:CD (SD) rat, male, 8 weeks
2 week oral gavage study, treated animal. Decreased body weight gain, proteinuria, increased plasma urea and ALT. Decreased plasma inorganic phosphorus. Distension of duodenum.
On low power, changes in the Brunner’s glands can be seen. On higher power, there is degeneration and necrosis of the Brunner’s glands with an acute inflammatory cell infiltrate and necrosis of the muscularis. There is a lymphocytic infiltrate in the overlying mucosa but villi appear normal.
A panel member commented that the changes seen here are characteristic for VEGF 1, 2 or mixed tyrosine kinase inhibitors. VEGF is expressed in certain areas of the gastro-intestinal tract, including Brunner’s glands (Hagedorn et al., Exp Tox Path, 2005, 57, 149-159). The changes seen may be a result in alterations in vascular permeability and/or secretory activity.
